I've had Enterprise running successfully for this DB since July. I have it on multiple machines. Today without any configuration changes Enterprise reports the following error message from all machines: Error No. 1 Could not connect to the tunneling URL. I have called the tunneling script directly with a web browser and it correctly returns a page. I attempted to watch the outgoing request with a packet sniffer and it appears Enterprise is not even sending an outgoing request before reporting failure. McAfee reports Enterprise has full access permission and Windows firewall is disabled. What is wrong?

all machines! Then it is very unlikely that it an issue on those client machines. Rather on the server side or on some transmission gear. Do you have an option to try another server. If not I could give you access to one.

You are PERFECTLY sure that the URL entered in SQLyog and the browser is the same?

Certain! I copied and pasted the URL from the SQLyog connect box into the browser.

The problem is on the clients. I loaded the software onto another machine and it works from there. All three machines are on the same LAN. There have been no manual configuration changes to either of the machines that won't connect. Windows firewall is disabled and McAfee reports full access. I tried with McAfee disabled and still the same.

      What could be wrong?

      Any 'automatic updates' configured?

If you have 'Systems Restore' enabled you can try returning to a restore point a few days back.
